چکیده

Bacteriophages insert their DNA into host chromosomes either through transposition (as in phage Mu) or through site-specific recombination (as in phage X). Whereas Mu can insert almost anywhere along the chromosome, X has a single highly preferred chromosomal site. Certain plasmids also insert into chromosomes by site-specific recombination. The site-specific recombinases used generally belong to the integrase family, whose members show some sequence homology and conservation of reaction mechanism, indicating descent from a common ancestor (2, 8, 12, 19). This minireview examines some chromosomal sites with known nucleotide sequences. The purpose is not to prepare an exhaustive catalog but to look for common trends. The focus is on chromosomal (attB) sites, not on the phage (attP) sites. Nevertheless, interpretation requires compari-

Active segregation by the Bacillus subtilis partitioning system in Escherichia coli.

Bacterial genes required for proper partitioning consist of two transacting genes that encode proteins and a cis-acting gene that functions like a centromere. Plasmids actively partitioning by means of these genes migrate from midcell to the cell quarters and are tethered to these sites until the cells divide. Previously the partitioning genes were mainly found on plasmids and phages in Escheri...

Cargo capacity of phages and plasmids and other factors influencing horizontal transfers of prokaryote transposable elements

Horizontal transfer of transposable elements (TEs) plays a key role in prokaryote genome evolution. Most TEs do not encode the enzymatic machinery allowing them to transfer between host cells and it is widely assumed in the literature that horizontal transfer of prokaryote TEs is mediated by other mobile genetic elements such as phages and plasmids. In a recent study, we have shown that phages ...

Generalized transduction in Streptomyces coelicolor.

We report the isolation of generalized transducing phages for Streptomyces species able to transduce chromosomal markers or plasmids between derivatives of Streptomyces coelicolor, the principal genetic model system for this important bacterial genus. We describe four apparently distinct phages (DAH2, DAH4, DAH5, and DAH6) that are capable of transducing multiple chromosomal markers at frequenc...

A Biobrick Library for Cloning Custom Eukaryotic Plasmids

Researchers often require customised variations of plasmids that are not commercially available. Here we demonstrate the applicability and versatility of standard synthetic biological parts (biobricks) to build custom plasmids. For this purpose we have built a collection of 52 parts that include multiple cloning sites (MCS) and common protein tags, protein reporters and selection markers, among...

The partitioned Rhizobium etli genome: genetic and metabolic redundancy in seven interacting replicons.

We report the complete 6,530,228-bp genome sequence of the symbiotic nitrogen fixing bacterium Rhizobium etli. Six large plasmids comprise one-third of the total genome size. The chromosome encodes most functions necessary for cell growth, whereas few essential genes or complete metabolic pathways are located in plasmids. Chromosomal synteny is disrupted by genes related to insertion sequences,...
